쥐성상세포와 해마절편에서 베타아밀로이드에 의한 인터루킨6 발현

Production of cytokines following overactivation of astrocytes and microglia is thought to be one mechanism that contributes to development of Alzheimer's disease (AD). Here we investigated conditions under which interleukin-6 (IL-6) expression is activated in astrocytes and hippocampal slices of the rat. Glutamate activated IL-6 expression in hippocampal slices and astrocytes. GABA did not affect glutamate-induced IL-6 expression, suggesting that the glutamate effect is not triggered by neuronal depolarization. Taurine, however, blocked glutamate-induced IL-6 expression. LPS also increased expression of IL-6. Taurine, unlike in the case of glutamate-induced increase of IL-6 expression, did not block LPS-induced IL-6 expression. These results indicate that glutamate and LPS act through different pathways to induce IL-6 expression. Beta-amyloid also enhanced IL-6 expression. The present results suggest that IL-6 expression can be induced by various factors and through several different pathways. Delineating interactions among different factors and different pathways may help understand pathogenesis of AD.
