Cell therapy has the potential for providing new treatments. Mesenchymal stem cells (MSCs) are important tools for cell therapy because of their high proliferation and differentiation ability as well as immunosuppressive properties. MSCs are commonly cultured as monolayers, but there are indications that they lose cell-specific properties with time in vitro. Additionally, recent publications demonstrated that the culture of MSCs as a spheroid form for short times improved their therapeutic potential by increased expression of genes such as CXCR4 to promote adhesion to endothelial cells or IL-24 that has tumor suppressing properties. In this study, we used periosterium-derived progenitor cells (PDPCs) that is a kind of MSCs, and developed a new culture system for spheres. Spheres could be passaged at least three times. After enzymatic dissociation, spheres could be reverted to monolayer cultures in flasks and they expressed MSCs-specific surface markers. In conclusion, this study can be utilized for the scale-up suspension culture of the PDPCs.
