A simple, rapid and reliable UPLC-MS/MS method for the simunltaneous determination of aminoglycoside antibiotics in human plasma

A simple, rapid and reliable ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was developed to determine simultaneously the aminoglycoside antibiotics (AMGs) amikacin, gentamicin, isepamicin, kanamycin, micronimicin, netilmicin, tobramycin, streptomycin in human plasma. Samples were prepared by a simple protein precipitation. The separation of AMGs was performed on a Kinetex HILIC column (50 × 2.1 mm, i.d., 1.7 μm) in gradient condition. The AMGs were detected by electrospray ionization mass spectrometry in positive ion mode with multiple reaction monitoring (MRM). The method was fully validated over the concentration range of 0.1 ~ 10 ug/mL for all AMGs. The precision and accuracy of seven AMGs were all below 15 % (CV) and no interferences from endogenous substances were found. The described UPLC-MS/MS method is suitable for therapeutic drug monitoring and for clinical pharmacokinetic studies of the AMGs.
