Effect of acidity on the hyperthermic enhancement of drug cytotoxicity.

The cytocidal effects of hyperthermia and many alkylating compounds have been known to increase when the environmental acidity is increased. However, little is know about the effects of acidity on the combination of hyperthermia and alkylating drugs. The present report describes the results of ongoing research in our laboratory on the effects of an acidic environment on the hyperthermia-induced increase in cytotoxicity of bleomycin and melphalan. The clonogenicity and apoptosis of tumor cells treated in vitro, as well as the growth delay of tumors in vivo, were the endpoints used in judging the effects of the various treatments.
