CapioCyte™ technology for high-sensitivity capture of circulating tumor cells: from nanoscale engineering to clinical validation

Circulating tumor cells (CTCs) enable the capture of tumor heterogeneity, serving as a potential biomarker for longitudinal monitoring of cancer progression. However, their inherently low abundance in peripheral blood has limited the performance of conventional isolation methods, thereby hindering the clinical translation of CTC-based diagnostic platforms. This review highlights the CapioCyte (TM) platform, a next-generation CTC isolation technology that combines biomimetic cell rolling with nanoengineered dendrimer surfaces to achieve markedly higher capture sensitivity than standard immunoisolation systems. We first outline the biological significance of CTCs and summarize current CTC-based liquid biopsy platforms in clinical or near-clinical use. We then discuss how the specific design of CapioCyte (TM), integrating E-selectin-mediated recruitment with multivalent antibody binding, addresses the limitations of existing approaches. Finally, we examine how high-yield CTC isolation enables downstream molecular and functional analyses that can guide treatment selection and improve patient outcomes. Although still in development, CapioCyte (TM) shows strong potential to advance precision oncology through highly sensitive CTC capture and clinically relevant applications.
