Production of biopharmaceuticals from mammalian cell culture has been steadily increased for the treatment of human diseases. To meet these demands, high productivity is essential for biopharmaceuticals produced in mammalian cell culture. Chinese hamster ovary (CHO) cells are the cell line used widely for the industrial production of various recombinant therapeutic proteins. Storage-protein 2 (SP2) is a soluble protein derived from silkworm hemolymph. Molecular weight of SP2 is 70 kDa and amino acid sequence is already confirmed. In addition, it has been known to inhibit apoptosis as well as cell aggregation and to inactivate reactive oxygen species in cell cultures. In this study, effects of SP2 on the transfected CHO cells producing albumin-erythropoietin (Alb-EPO) were investigated. As a result, it was found that the viable cell density and viability could be enhanced with SP2. Furthermore, reduction of cell aggregation was also observed. Especially, decreasing cell aggregation caused the reduction of apoptosis. In conclusion, SP2 showed positive effects on CHO cell cultures.
