Human Papillomavirus DNA in various cervical lesions of Korean Women: Detection and typing by PCR-RFLP and ologonucleotide microarray

The association between cervical carcinoma and human papilloma virus (HPV) is now well established. To estimate the extent of infection with common HPVs in various cervical lesions among Korean women, we have examined the 464 cervical scrapes of various cervical lesions. Ditection and typing of HPVs were done by polymerase chain reaction (PCR) amplified with consensus primers followed by restriction enzyme digestion and oligonucleotide microarray. The prevalence of total HPV infection in patients were gradually increased with the increased severity of the disease; HPV DNA was detected in 31.8%, 33.3%, 54.5%, 70%, 70.7% and 86.3% of the cervicitis, condyloma acuminatum, CIN I, CIN II, CIN III and invasive squamous cell carcinoma, respectively. HPV typing in 79 CINs revealed prevalence of HPV 16 in 17 cases followed by HPV 33, 31, and 58, 52, 35 and 18. HPV typing in 73 invasive squamous cell carcinoma revealed a prevalence of HPV 16 in 18 cases followed by HPV 33, 31, 58, 52 and 18. The prevalence of HPV 16 was significantly increased in invasive carcinoma when compared to CIN. The other HPV types appeared to be more prevalent in CIN than invasive carcinoma. In conclusion, these findings suggests that HPV 18 is less frequently associated in squamous cervical neoplasia and the so called 'intermediate-risk HPVs' are more prevalent in both CIN and invasive squamous cell carcinomas, however, this viruses appear to have less oncogenic potential than high-risk viruses.
