Arsenic trioxide reduces tumor blood flow, inhibits endothelial cell growth and selectively kills hypoxic cells

We have investigated the effect of an anti-leukemic drug, Arsenic Trioxide(ATO), against experimental murine solid tumors as well as against endothelial cells and tumor cells in culture. There was obvious macroscopic vascular damage in the central region of 7-9mm subcutaneous FSall tumors grown in C3H mice by 2h after an injection of 10mg/kg which persisted until at least 24h after injection. Tumor blood flow, measured by Rb uptake, was reduced to 43% of control level by 30 min after injection and decreased to a low of 17% of control at 16h after injection.
