Chondrogenic differentiation of mouse embryonic stem cell in vitro
초록
In vitro systems used to study chondrogenesis are primary culture of adult or embryonic cartilage tissue and cell lines of mesenchymal origin of mesenchymal stem cells derived form bone marrow.l However, these cell systems are limited for studies of early differentiation because they represent already determined and differentiated cells. An alternative approach is offered by the differentiation of pluripotnent mouse ES cells. ES cells have the capacity to differentiate into cell s of all three primary gerlayers due to their origin from the inner cell mass of early embryos. Furthermore, members of the transforming growth factor-b family have the potency to influence deveolpment of cartilage and bone, Our object was to set-up the techanique for chondrogenic differentiation of ES cells. We demonstrated that mouse ES cells differentiated in vitro into chondroyctes. And we showed that members of the TGF-b growth factor family modulate ES cell-derived chodnrogenic differentiation in a time-dependent manner.
