Molecular cloning of novel organic anion transporter (mOATbv) from mouse aorta

Solute carrier family (SLC) including organic cation transporters, organic anion transporters and carnitine transporters play an important role for movement of organic ionic compounds and zwiter ion in cells. It is well known that several transporter proteins act as blood brain barrier. In this connection, we try to search organic transporter in cDNA library prepared from mouse blood vessel using human OAT4 complementary DNA (cDNA) as a query. Employing the hybridization cloning method, a cDNA was isolated. The mOATpv cDNA consists of 1997 base pairs that encode 553-amino acid residue protein. Hydropathy analysis suggested that OATpv possess 12 putative membrane-spanning domains the same as other reported mouse OAT isoforms. When we aligned the amino acid sequence of the known mouse OATs and mOATpv to generate a dendrogram delineating their phylogeny, mOATpv showed mOAT1 with 41%, mOAT2 with 37%, mOAT3 with 41%, mOAT5 63%, mOAT6 85% and rat UST1 with 63%. The northern blot analysis using total RNA of blood vessel showed about 2.1 kb message band. The functional characterization will be necessary. From these results, mOATpv may give some understanding on the movement of organic compound including metabolites and drugs.
