Comparison of protein expression profiles between the wild type Haematococcus pluvialis and their mutants
초록
The green alga Haematococcus pluvialis accumulates large amounts of the astaxanthin (3,3…-dihydroxy-??carotene-4,4…-dione) when exposed to stress condition especially under high light intensities, salt stress, etc. Astaxanthin is a ketocarotenoid pigment with potential effects of antioxidant. In order to obtain the better producer of astaxanthin, H. pluvialis cells were mutated with colchicines treatment and U.V irradiation. After a series of selection, about a promising mutant cell with 1.6-fold higher contents of total carotenoid per cell than that of the wild type strain was identified. In addition, another mutant cell line that showed a faster growth rate was selected the protein expression patterns of mutated H. pluvialis cell lines were compared with that of wild type strain using native-PAGE, SDS-PAGE and 2-D electrophoresis. The expressed protein set of mutant cells was somewhat different from that of wild type cells. In native- and SDS-PAGE, Moreover, 2-D gel analysis revealed distinctly different between the mutant and wild type cells.
