알레르기비염에서 항원유발반응 후 비즙 내 일산화질소 및 peroxynitrite의 형성
초록
Background. Allergic reaction is mediated by released histamine or various chemical mediators which are released upon stimulation by specific antigen and allergic reaction can induce vasodilation and extravasation of plasma. Challenge with specific antigen can elicit both early and late phase responses of allergic reaction. The presence of nitric oxide(NO) in nasal secretion is well documented and recent evidence has shown that NO level in nasal secretion is increased during allergic reaction. However the role of this molecule in the pathophysiology of allergic rhinitis is still poorly understood. Peroxynitrite, reactive metabolite produced from NO, is potent oxidizing agent that nitrates tyrosine residues present in the proteins of nasal fluid and cause nitrotyrosine formation, and produces deleterious effects on protein function. Objectives. In this study, we atempted to clarify whether NO production is increased in the nasal secretion of allergic rhinitis patient during early or late phase of allergic reaction induced by allergen. We also investigated whether the albumin and NO concentration in nasal secretion is correlated because albumin release is used as an index of increased vascular permeability, and NO is a strong inducer of vasodilation and plasma extravasation. We also determined the formation of nitrotyrosine, as an index of cellular toxicity by peroxynitrite formation, during early and late phase of allergic reaction. Materials and Methods. Nasal secretion samples were obtained from twenty three patients showing allergic rhinitis to Dermatophagoides pteronyssinus using both the lavage methods (n=10) and the filter paper absorption method(n=13). The nasal secretion samples were collected three times; before the allergen challenge(baseline), 30 minutes after(early phase), and 8 hours after(late phase) the allergen challenge.
