MMS resistant mutations isolation: gene search against toxicants
초록
Environmental DNA damaging agents induce cellular toxicology causing cell cycle alteration, metabolic changes, mutation and even cell death.With such, cells respond to damaging agents to protect by the elimination of toxic chemicals and recovery from damage by camaged cellular components repair. In our previous studies, we presented the effects of Methyl methynesulfonate (MMS) on cell survival and mutagenesis.We also observed that the damage of DNA repair genes led to a decreased yeast lifespan.As an attempt to isolate genes that affect MMS sensitivity, yeast insertional deletion librarywas used to idenify the genes that might increase lifespan despite the presence of damaged DNA. As the results, several genes, previously known to be unrelated to MMS sensitivity, was identified to increase or decrease MMS sensitivity. Currently, by investigating the characteristics of targeted-gene deletin mutants, we are confirming the effects of each gene on MMS sensitivity.
