The TGF-β-inducible early gene 1 regulate allergic skin inflammation by phthalic anhydride

Recently, several studies revealed that TGF-β-inducible early gene 1 (TIEG1) is closely related with immune system. It has been shown that TIEG1 deficiency causes defective in inhibiting airway inflammation and results in excess of Th2-mediated allergic response. However no trials have ever been conducted to elucidate the role of TIEG in allergic skin inflammation. Thus, the aim of the current study was to investigate the role of TIEG1 in allergic skin inflammation induced by phthalic anhydride (PA). To accomplish this, 1, 5 and 10% PA solution as chemical allergen was repeatedly spread on the ear’s dorsum in TIEG1-/- and its C57BL/6 mice for 3 weeks. After treatment, the allergic and immune-related factors were detected in the ear tissue, lymph node, and serum, respectively. The ear thickness, inflammatory cell infiltration, and Myeloperoxidase activity, all of which indicating degree of allergic skin inflammation was significantly (P<0.05) increased in the TIEG1-/- mice compared with C57BL/6 mice. Furthermore, inflammatory cytokines in inflamed tissue and concentration of Th2 type immunoglobulins (IgE and IgG1) in serum was significantly (P<0.05) high in the TIEG-/- mice. These findings suggest that the TIEG1 regulates allergen-induced skin inflammation. Thus, the identification of TIEG1 may provide new insights for additional therapeutic targets of allergic skin inflammation such as atopic dermatitis
