Objectives: We hypothesized that combined treatment with olanzapine (OLZ) and green tea (GT) extracts or EGCG differs from olanzapine monotherapy in the aspect of the effects on body weight and serum levels of lipid markers, nitric oxide and glucose. Methods: ICR male mice weighting average 30.12±1.03g at the beginning of the experiment were used. The animals were housed five per cage and had free access to food and water. OLZ and GT groups were tested to observe their effects on the obesity (weight and plasma levels of total cholesterol, triglyceride and glucose). OLZ-GT and OLZ-EGCG groups were to analyze the effects of green tea and EGCG on drug-induced obesity in OLZ-treated group, respectively. Results : The average weight of animals was 30.12±1.03g, prior to the administration. The chronic treatment of OLZ significantly increased the average body weight with approximately 2-fold rates (22.7-39.6%, 29.2±3.96g-40.76±3.10g) than that of control (10.1-18.8%, 30.1±4.18g-35.77±2.41g) when the average weights estimated every 10 days were compared to those of the control. In presence of green tea or EGCG, the drug-induced increasing rates of the body weight were dramatically decreased to the even less (-7.1-5.03%, 31.1±5.48g-32.56±2.91g) than the control or similar (7.6-18.0%, 29.0±4.18g-34.21±3.71g) with the control rates, respectively. In the OLZ-treated group, the plasma levels of TG, nitric oxide (NOx) and glucose (Gl) were significantly increased, whereas not changed in those of the three lipid markers, TC, HDL, and LDL. The green tea or EGCG prevented the increase of OLZ-induced plasma level of TG, NOx and Gl. Conclusion : These results suggest that green tea or EGCG can protect the olanzapine-induced weight gain, hyperglycemia and hypertriglycemia.
