Quantification of Fibrillation Potentials in relation to the Severity of Injury and Change of Temperature in Sciatic Nerve Injured Rats

Extremes of temperature profoundly influence to the normal physiology of nerve and muscle. Although compression injury of the peripheral nerve for temperature change has been studied, the correlation between the severity of nerve injury and quantification of fibrillation potentials for temperature change has not been fully investigated. This study was performed to evaluate the patterns of fibrillation potential in relation to the severity of injury, and to investigate the effect of temperature change. We used 10 male Sprague-Dawley rats. The both sciatic nerves were compressed in 3 different degrees with haemostatic forceps. The experimental group was divided into 4 groups according to the intensity and duration of nerve compression: group 1, first degree compression for 5 seconds: and group 2, first degree for 30 seconds; group 3, third degree for 5 seconds; and group 4, third degree for 30 seconds. The electromyogram of the both gastrocnemius muscles were recorded in experimental rats at the post-injury 5 days with a needle electrode and digital thermometer probe were inserted. The calf muscle of the rat was warmed with fan heater until the temperature reached to 42 ℃ and cooled with fan cooler until to 15 ℃. We counted the number of fibrillation potentials in gastrocnemius muscle in a wide range of temperature change. The quantities of fibrillation potential were highly correlated with the temperature change in large damaged group. These results may provide the basic data to clarify the fibrillation potentials in relation to the severity of injury, and to help establishing adequate temperature after peripheral nerve damage.
