Primed human clonal mesenchymal stem cells enhance therapeutic efficacy in a murine model of atopic dermatitis

Mesenchymal stem cell (MSC)-based cell therapy has received great interest in regenerative medicine and therapies for immune diseases because of their remarkable anti-inflammatory, immunomodulatory, and regenerative properties, which involve both paracrine and cell-to-cell contact mechanisms. However, most of the first-generation approaches displayed heterogeneous clinical outcomes regarding efficacy because of different sources, donors, and variability of culturing methods. Thus, in recent years, priming approaches to improve therapeutic efficacy have been investigated and we found a new combination of soluble factors that enhance the therapeutic effect on atopic dermatitis animal models. Adding IFN-alpha and Vitamin B6 to the previously well-known stimulus TNF-alpha and IFN-gamma combination increases immune modulatory factors such as ICOSL, TSG6, PD-L1, Galectin 9, and IL-1 receptor antagonists. In addition, the primed MSCs could increase Foxp3+ IL-10+ regulatory CD4+ T cells. We think that the new priming approaches might help improve the clinical therapeutic efficacy of MSCs.
