Modeling of Aminotransferase Structure by Sequence Similarity

초록

ω-Aminotransferase reversibly catalyzes the transamination reaction for particular substrates with pyridoxal 5’-phosphate (PLP as a cofactor. Theoretical structure of ω-aminotransferase was constructed by GeneFold moldule using the sequence similarity. 3D-structure of protein was built from both sequence and structure information to measure sequence-structure compatibility using three different scoring functions. To elucidate the multiple substrate recognition mechanism. We constructed complex structure of ω-aminotransferase bound to PLP with LYS-285. PLP is located in the active site by specific hydrogen bonding with aspartate and arginine residues.
