저강도 초음파 자극이 alginate 배양에서중간엽 줄기세포의 연골세포 분화에 미치는 영향
초록
Many growth factors, cytokines and mechanical stimulation affect chondrogenic differentiation of mesenchymal stem cells (MSC), that are capable of differentiate into osteoblast, chondrocytes, adipocytes, and myocytes. However, the mechanismas of how the factors affect chondrogenic differentiation remain unclear. To date, three dimensional culture and TGF-b are known as tow of the essential factor for chondrogenesis. Low-intensity ultrasound (LIUS) treatment enhances fracture healing via increase of chondrogenesis suggesting its possible funcion in chondrogenesis of MSC. The object of this study was to examine the effect of LIUS on chondrogenic differentiation of rabbit MSCs. Ultrasound stimulation on rabbit MSCs cultures in a 3-dimensional environment of alginate beads and serum free defined media with TGF-b1 or 3 increase 1) the cell viability by 30%, 2) expression of the collagen type II, and 3) the TIMP-2 expression. The results indicate that LIUS stimulated chondrogenic differentiation of rMSC suggesting that the noninvasive ultrasound fuctions as a chondrogenic mechanical stimulus.
