Effects of Mixing Time in Wave Bioreactor on the Production of hCTLA4Ig in Transgenic Rice Cell Cultures

Plant cell culture has been used as an attractive tool for the production of various recombinant proteins such as antibodies, enzymes, vaccines and blood factors [1]. Disposable bioreactors were initially used only in research and development, but they are increasingly used in commercial manufacturing operations during the last 5 years [2]. In this study, effects of mixing time on cell growth and hCTLA4Ig production were investigated using CultiBag RM 20 L (Sartorius Stedim Biotech) in transgenic rice cell cultures. Two mixing times were tested. Those were 15 sec (rocking rate: 25 rocks/min, angle 8°, KLa: 4.5 h-1), and 8 sec (rocking rate: 30 rocks/min, angle 10°, KLa: 5.8 h-1). Both conditions were applied to bubble-free air sparging (0.3 vvm) with periodic pure O2, and the working volume was 8 L. Maximum dry cell weight with mixing time 15 sec was 1.1-fold higher and maximum hCTLA4Ig production was 2.5-fold higher than those with 8 sec. In conclusion, it is clear that mixing time is important in wave bioreactors for transgenic plant cell cultures.
