One-Step Evaluation of Doxorubicin Toxicity Through Concentration Gradient Generator Integrating Breast Cancer-on-a-Chip

Conventional drug screening platforms face limitations such as single cell line analysis, slow processing, unstable fluid flow, and frequent gradient adjustments, highlighting the requirement for high-throughput alternatives. In this study, a breast cancer-on-a-chip platform integrated with a concentration gradient generator was introduced, enabling rapid and reproducible anticancer drug evaluations. Computational fluid dynamics (CFD) ensured stable gradient generation through convection and diffusion mechanisms. The platform successfully mimicked a triple-negative breast cancer (TNBC) microenvironment by co-culturing MDA-MB-231 and MCF-10A cell lines within the chip. High-throughput screening of doxorubicin (DOX) was conducted, demonstrating 82% survival of MCF-10A cells and 50% death of MDA-MB-231 cells at 18.25 mu M DOX. Therefore, the newly designed platform offers precise evaluation of drug efficacy under physiologically relevant conditions and provides valuable insights into effective anticancer drug concentrations for clinical applications.
