Adhesion characteristics of Trp-Lys-Tyr-Met-Val-D-Met (WKYMVm) and human formyl peptide receptor (hFPR) using atomic force microscopy (AFM)

Atomic force microscopy (AFM) has been used to investigate the dissociation of a ligand-receptor; its direct measurement can make the analysis results time-independent, and can be performed within a few minutes. Trp-Lys-Tyr-Met-Val-D-Met (WKYMVm), the robust neutrophil chemo-attractant, is a synthesized agonist for formyl peptide receptor (FPR), which triggers PLC-mediated signal transduction. The kinetic and thermodynamic properties of WKYMVm-FPR interaction have been investigated by conventional methods but not the adhesion force of WKYMVm-FPR. In this research, we investigated the adhesion force of WKYMVm-FPR by using AFM and suggested its usefulness in analyzing its kinetic and thermodynamic characteristics.
