Transgenic rice cell suspension cultures were used as a production system for recombinant proteins. High-density cultivation is important for the effective production system. Therefore, the shortening of the growth period is required to reach high cell density. Amino acid (AA) medium is consisted of basal salts, amino acid mixtures, and growth regulators. However, AA medium has low concentration of nitrogen sources. Therefore, additional supplementation of nitrogen source to AA medium is necessary for the better growth of transgenic rice cells. In this study, the effects of peptide mixtures such as casein enzymatic hydrolysate and phytone peptone during the growth phase were investigated. Various concentrations of each peptide mixture at 1, 2, 3% (w/v) were tested in 100-mL Erlenmeyer flasks. Cell mass and specific growth rate were measured in growth period, respectively. The optimum concentration of casein enzymatic hydrolysate and phytone peptone showed an increase of biomass as well as fast growth. In conclusion, supplement of peptide mixtures as an alternative nitrogen source can enhance the growth rate and increase cell mass in transgenic rice suspension cell cultures.
