Comparison of growth profiles for the production of acid-alpha glucosidase between wild type and gnt1 mutant transgenic rice cells

Acid-alpha glucosidase (GAA) is used in enzyme replacement therapy for Pompe disease. GAA is delivered to lysosome by M6P receptor-mediated pathway. In plant cells, knock-out of gnt1 gene could express glycoproteins with high-mannose glycans. However, absence of gnt1 gene in plant cells exhibits physiological differences such as salt sensitivity and cytokinin insensitivity. In this study, GAA was produced in wild type and gnt1 mutant transgenic Oryza sativa suspension cell lines using three kinds of media (N6, B5, AA). In the case of wild type, there were no significant differences in dry cell weight (DCW) in all kinds of media. However, viability was the highest in N6 medium. On the other hands, maximum DCW of gnt1 mutant was 12.72 g at day 8 and the highest cell viability was also obtained in N6 media. In addition, browning of rice cells was observed in AA and B5 medium, in which protease level was found to be high. In conclusion, for the cultivation of gnt1 mutant line, optimization of culture condition is required.
