Crohn's disease Animal Model in Mini-pig

Crohn’s disease is a chronic inflammation intestine disorder targeted at the entire digestive organs, namely from mouth to anus. It is usually founded along the large intestine, in which it is scattered distribution pattern. There are many therapeutic method and drugs developed for this disease with an effective way. But, there is still many hurdle to handle it in terms of the reason of disease, full recovery and appropriate animal models etc. Mini-pig is known to be a most similar animal to human based on its size, anatomy as well as physiology. This is the reason why we chose swine animal model for the efficacy test of Crohns’ disease. We use four mini-pigs for this experiment with two group, mock control and DNBS treated. We injected 10ml/kg of 30% DNBS in ethanol into colon by endoscope. Once we pour off this chemical onto colon, we try to keep this solution at the colon of 1 mini-pig for 2 min. The other is flow out the solution right after it pour off from the endoscope. After 4 days, we checked the colon length as well as its anatomy. There is no sings specifically and looks fine at mock control pig. But DNBS treated group showed some signs of depression as well as round nodules (2~3mm) at distal bloated large intestines’ serosal surface, in which some region of the colon appeared dark colored and hardened at inside. And also, the colon length is increase in DNBS treated group compared to the mock control. We found that the pig treated DNBS has a similar symptoms of Crohn’s disease and is useable animal model.
