To separate ibuprofen which is chiral material, we used a chromatographic column packed with chiral stationary phase. Mobile phase used were n-hexane and 2-propanol(IPA) with the chemical buffers of trifluoroacetic acid(TFA) or acetic acid(AA). The flow rate of mobile phase was 1.0㎖/min and injection volumn was 5㎕. The resolutions of ibuprofen enantiomers were investigated with the composition of mobile phase on a chiralcel OD-H column. From the experimental results, as the amounts of IPA were increased, the resolution of enantiomers was worse because of shorter retention times. Compared to TFA, in the chemical buffer of AA, the shape of peaks was sharper. It was also observed that as the more amount of AA were added, the longer retention factors caused the better resolutions. The most favorable resolution to separate the two enantiomers was obtained at 100/1/0.12, n-hexane, IPA and AA (vol.fraction).
