ISOLATION OF A MARINE BACTERIOPHAGE INFECTING STRAIN IMCC1322 OF THE SAR116 CLADE AND ITS COMPLETE GENOME SEQUENCE

The SAR116 clade in the Alphaproteobacteria comprises approximately 5% of the total bacterial community in the ocean surface. Here we report isolation and the genome sequence of a bacteriophage that infects a representative strain of the SAR116 clade, strain IMCC1322. IMCC1322phage#1 was recovered from a surface seawater sample collected from the East Sea where the host was isolated, and the viral genome was analyzed using whole genome shot gun sequencing. IMCC1322phage#1 had doublestranded DNA genome of 55,282 bp and it harbored modules of genes for replication machinery and structural proteins. In addition the genome contained putative genes for Rieske (2Fe-2S) domain protein, phosphoglycerate mutase, L-gulono-gamma-lactone oxidase, and transcriptional regulators. Based on DNA polymerase phylogeny and transmission electron microscopy, it is suggested that IMCC1322phage#1 belong to an unassigned Podoviridae group, distantly related to other marine bacteriophage isolates such as Roseophages SIO1, DSS3phi2 and EE36phi1.
