The aim of this study was to improve microalgal biomass and fatty acid (FA) productivities using the newly isolated freshwater microalga Micratinium sp. KGP-12 via sequential optimization of culture conditions. First, the appropriate temperature and light intensity for Micratinium sp. were determined as 30℃ and 300 μmol/m2/s, respectively, using a high throughput photobioreactor. The Box-Behnken experimental design technique was adopted to optimize the culture medium by adjusting the N, P, and Mg concentrations in BG-11. The biomass and fatty acid productivities from optimized BG-11 increased by up to 45%, compared to those from standard BG-11 in 0.5 L bubble column photobioreactors. Analysis of the FA composition did not show any differences in either composition or content. When the cells were subjected to N- and/or P-deficient media to increase cellular FA content, N deficiency groups (N-P+ and N-P-) presented a fast increase in FA content from 8.8% to 13.6% on day 2, whereas that of P-deficient medium was only 9.8%. Overall, the results indicated that biomass and FA productivities could be effectively enhanced via the optimization of culture medium and appropriate nutrient supply strategies. © 2025, The Korean Society for Microbiology and Biotechnology.
