Perfusion Culture of Transgenic Nicotiana tabacum Suspension Cells for the Production of Human Granulocyte Macrophage-Colony Stimulating Factor (hGM-CSF)

Transgenic Nicotiana tabacum suspension cells producing recombinant human granulocyte macrophage-colony stimulating factor (hGM-CSF) were cultured in both batch and perfusion modes in a 5-L stirred tank bioreactor. During the perfusion culture for improving productivity, cell density could be obtained up to 22.4 g/L which was 1.4-fold higher value compared to 16.1 g/L of batch culture. The productivity of hGM-CSF could be maintained at 3 mg/L/day until the end of culture with continuous addition of fresh medium. As a result, 1.1 mg of extracellular hGM-CSF was accumulated in 26 L of spent medium at the end of 16-day operation.
