세포부착 융합 단백질의 제조합 유전공학
초록
We have previously reported the synergistic effect of fibronectin and fibroblast growth factor 2 (FGF-2) on osteoblast cell adhesion through extracellular signal-regulated kinase (ERK) pathway (BBRC 2002 Jul 26; 295(4):898-902). Here we describe engineering of a fusion protein containing fibronectin fragment (FNIII9-10) connected to the COOH-terminus of FGF-2. Purified FGF2-FNIII9-10 fusion protein exhibited significant increased cell adhesion and proliferation of human osteoblast-like MG63 cells compared with FNII9-10 or FGF-2 alone (p<0.05). This fusion protein design may thus serve as a general route for immorbilizing a variety of bioactive sequences to biomaterials surfaces for applications in biomaterial coatings, tissue engineering, and dental implant.
