초록
The dynamic change of intracellular Ca2+ concentration ([Ca2+]i) is a fundamental neuronal process involved in the induction of long-term depression (LTD), neurotransmission, gene expression, cell death, neuronal development and neuronal migration in Purkinje neurons. Purkinje neurons have powerful systems to modulate [Ca2+]i, such as Ca2+-binding proteins, intracellular stores, Ca2+-ATPase and Na+-Ca2+ exchanger (NCX). Cerebellar LTD is expressed as a functional downregulation of AMPA receptors associated with ion channels. The physiological roles of NCX in Purkinje neurons are not well understood, especially in cerebellar LTD induction. Thus, in this study, we investigated using cellular Ca2+ imaging technique the physiological roles of NCX in modulation of Ca2+ entry via AMPA receptors of rat cerebellar Purkinje neurons. In addition, there have been reports that NCX is associated with caveolin-3 in cardiac sarcolemmal vesicles. And recent studies have also provided evidences that caveolins are expressed in brain. But the functional significance of caveolins remains unknown. Therefore, we also studied the interaction between NCX and caveolins in C6 glioma cells.
