Seperation of Acanthoside-D from Acanthopanax Senticosus with Supercritical Fluid Extraction

The feed samples were obtained from 120g powder of Acanthopanax senticosus. They were extracted using supercritical carbon dioxide at the temperature of 50 oC and the pressure of 200, 250, and 300 bar and the CO2 flow-rate of 200 l/hr. The water content of 0.5, 1.0, 1.5, and 3.0 ml/mg were maintained at 50 oC, 300 bar, and 200 l/hr. The supercritical fluid extracts were partitioned with water and hexane to perform the solvent extraction. The resulting solution was analyzed with a chromatographic column (3.9?e300 mm, 15 ?, Lichrospher 100RP -18). The mobile phase composition for resolving Acanthoside-D from the extract was water/acetonitrile/methanol, 80/14/6 vol.%. The flow rate of mobile phase was 1 ml/min and UV wavelength was fixed at 210 nm. In an experimental run without the additives of water, the yield of Acanthoside-D was larger at higher pressure. But, the residual phase in the supercritical extractor showed the higher amount of Acanthoside-D with the water additives, because Acanthoside-D was a moderately polar material. The phase contained polar materials mainly of water, where the concentration of Acanthoside-D reached up to 70 mg/ml. The sample was purified by open tubular chromatography and further isolated by high performance liquid chromatography.
